【分离原理】一般血清比重约1.030左右、血块比重约1.092左右、本公司分离胶比重为1.045~1.065之间。分离胶具有良好的触变性能和隔离性能。当分离胶与凝固后的血液在同一试管中离心时,在离心力的作用下,分离胶分子间氢键断开转化为低粘度流体,由于其比重居于血球和血清之间,在离心过程中分离胶便逐步堆积在血清和血块之间。当外在离心力消失后,分子间的氢键重新缔合形成网状结构,分离胶重新成为高粘度的胶体,形成致密的隔离层将血清和血块隔开。
【使用方法】
1.加胶:可采用机器或者手工方式加料;
(1)用自动、半自动加胶机加胶时,首先将分离胶加入机器料斗中,待胶体流平,盖好加料斗,即可开始加料。在冬季气温较低时,有条件的情况下可适当加温,采用油浴、水浴或者电加热的方式均可,温度控制在30-60℃.
(2)手工加料时,将胶体注入加料器中,加料头置于试管底部,缓慢推进加料杆,随着胶体缓慢流入试管,提升加料头,加入足够胶体后,小心拿出加料器即可。
2.排气泡:采用自动加胶机,加胶后将加入胶体的试管直立放置8-12小时,然后密封抽真空,约有70%左右的试管不会出现气泡。手工加胶可能造成大量气体夹入,抽真空后会有大量气泡出现。出现气泡时,建议用离心机在800-1200g离心力,离心2-5min排出气体。排气泡需要的离心力及离心时间和加胶、胶体转移过程中夹入的空气多少有关,建议使用过程中依据实际情况进行调整。
离心排气泡可以在加胶后2h进行,也可以在抽真空后1h以后进行。分离胶和促凝剂、抗凝剂同时使用时,建议先加入分离胶,然后排气泡,再加入促凝剂或者抗凝剂,然后再抽真空。这样可以使分离胶对促凝剂、抗凝剂的影响最小。

5.离心条件:

(2)制备血浆:在采血完成后,颠倒混匀5-8次,抗凝充分后即可离心。离心条件同上。
6.灭菌:本公司分离胶分为两种,一种为普通分离胶,不能耐受钴60辐照灭菌;一种为可辐照分离胶,可采用钴60辐照灭菌。建议在8-40KGY剂量范围内灭菌。辐照剂量以初始菌落数确定。

【添加剂量】每支试管建议加入分离胶的量为0.8-1.2g(0.75-1.1ml)。对于直径16mm的塑料管,加胶量应该在1.0-1.2g以上。判断加胶量是否足够的标准是,正常离心后胶体(隔离层)厚度≥6mm。
【禁忌症】无。
【注意事项】
本公司分离胶为专利产品,成分独特,请不要和其它供应商产品混用,以免造成产品残次,给你造成不必要的损失;
分离胶的推荐温度使用范围是-20~+90℃。本品随着环境温度变化,粘度会出现较大变化。环境温度过低,可能会导致加胶困难。建议在环境温度低于15℃时,可水浴加热到60~70℃,降低粘度,提高加胶效率;
常温加胶时出现“拉丝”现象,可以适当提高加胶温度,或者以振动方式将丝震断;
本品经过高真空处理,胶体中基本没有夹杂空气,通常不建议在转移、加胶过程中进行搅拌,因为搅拌易导致胶体中夹入大量空气;
本品加入试管后,在真空状况下经过连续20天以上60℃的倒立试验,胶体平面流淌距离≤5mm。可以保证以集装箱方式在60℃下连续运输20天不会出现超过5mm的流淌,不会影响采血管的正常使用。即便如此,我们仍然建议,在长时间运输过程中,含分离胶的采血管不倒置;
