【分离原理】一般血清比重约1.030左右、血块比重约1.092左右、本公司分离胶比重为1.045～1.065之间。分离胶具有良好的触变性能和隔离性能。当分离胶与凝固后的血液在同一试管中离心时，在离心力的作用下，分离胶分子间氢键断开转化为低粘度流体，由于其比重居于血球和血清之间，在离心过程中分离胶便逐步堆积在血清和血块之间。当外在离心力消失后，分子间的氢键重新缔合形成网状结构，分离胶重新成为高粘度的胶体，形成致密的隔离层将血清和血块隔开。
【使用方法】
1.加胶：可采用机器或者手工方式加料；
（1）用自动、半自动加胶机加胶时，首先将分离胶加入机器料斗中，待胶体流平，盖好加料斗，即可开始加料。在冬季气温较低时，有条件的情况下可适当加温，采用油浴、水浴或者电加热的方式均可，温度控制在30-60℃.
（2）手工加料时，将胶体注入加料器中，加料头置于试管底部，缓慢推进加料杆，随着胶体缓慢流入试管，提升加料头，加入足够胶体后，小心拿出加料器即可。
2.排气泡：采用自动加胶机，加胶后将加入胶体的试管直立放置8-12小时，然后密封抽真空，约有70%左右的试管不会出现气泡。手工加胶可能造成大量气体夹入，抽真空后会有大量气泡出现。出现气泡时，建议用离心机在800-1200g离心力，离心2-5min排出气体。排气泡需要的离心力及离心时间和加胶、胶体转移过程中夹入的空气多少有关，建议使用过程中依据实际情况进行调整。
离心排气泡可以在加胶后2h进行，也可以在抽真空后1h以后进行。分离胶和促凝剂、抗凝剂同时使用时，建议先加入分离胶，然后排气泡，再加入促凝剂或者抗凝剂，然后再抽真空。这样可以使分离胶对促凝剂、抗凝剂的影响最小。
静置：无论是加胶过程，还是抽真空、离心过程，都会导致分离胶触变性暂时降低，建议在加胶、离

4.本品可和促凝剂、抗凝剂（EDTA盐、肝素盐）等配合使用。
5.离心条件：
（1）制备血清：在血液充分凝固后，在离心机上以1800-2200g离心力离心8-10min即可制备高质量的血清。
（2）制备血浆：在采血完成后，颠倒混匀5-8次，抗凝充分后即可离心。离心条件同上。
6.灭菌：本公司分离胶分为两种，一种为普通分离胶，不能耐受钴60辐照灭菌；一种为可辐照分离胶，可采用钴60辐照灭菌。建议在8-40KGY剂量范围内灭菌。辐照剂量以初始菌落数确定。

【添加剂量】每支试管建议加入分离胶的量为0.8-1.2g（0.75-1.1ml）。对于直径16mm的塑料管，加胶量应该在1.0-1.2g以上。判断加胶量是否足够的标准是，正常离心后胶体（隔离层）厚度≥6mm。
【禁忌症】无。
【注意事项】
本公司分离胶为专利产品，成分独特，请不要和其它供应商产品混用，以免造成产品残次，给你造成不必要的损失；
分离胶的推荐温度使用范围是-20～+90℃。本品随着环境温度变化，粘度会出现较大变化。环境温度过低，可能会导致加胶困难。建议在环境温度低于15℃时，可水浴加热到60～70℃，降低粘度，提高加胶效率；
常温加胶时出现“拉丝”现象，可以适当提高加胶温度，或者以振动方式将丝震断；
本品经过高真空处理，胶体中基本没有夹杂空气，通常不建议在转移、加胶过程中进行搅拌，因为搅拌易导致胶体中夹入大量空气；
本品加入试管后，在真空状况下经过连续20天以上60℃的倒立试验，胶体平面流淌距离≤5mm。可以保证以集装箱方式在60℃下连续运输20天不会出现超过5mm的流淌，不会影响采血管的正常使用。即便如此，我们仍然建议，在长时间运输过程中，含分离胶的采血管不倒置；
